چکیده

In the course of our studies on the role of complement in an t i se rum-media ted damage to allografts and xenografts of skin, we have observed that, whereas serum from C5 deficient mice fails, as expected, to lyse sensitized sheep erythrocytes (SRBC),I whole blood from these animals causes measurable lysis even when di luted 1:100 in saline. This lytic activity occurs only if the cells have reacted with ant ibody, and only if platelets and early acting components of complement (C) are present. It appears to involve mechanisms not previously described in the l i terature and it may be useful in s tudying the functions of platelets as well as in del ineat ing the cytotoxic potent ia l of antibody. We describe here the condit ions under which the p h e n o m e n o n occurs and we present evidence for the essential involvement of ant ibody, complement , and platelets.

Lysis of antibody-coated cells by platelets

Antibody-coated erythrocytes are lysed by murine C5- whole blood but not by plasma separated from such blood. The lytic activity has been shown to derive from platelets that attach to sensitized cells probably through membrane receptors for C3b. Whole blood or platelet-rich plasma (prp) obtained from mice that have been treated with purified cobra venom factor has little or no activity unless i...

AET-treated platelets: their usefulness for platelet antibody detection and an examination of their altered sensitivity to immune lysis.

2-Aminoethylisothiouronium bromide (AET) increases the sensitivity of blood cells to complement-mediated immune lysis. We compared the sensitivities of untreated or AET-treated platelets to immune lysis induced by different types of platelet antibody in the 51Cr platelet lysis test. AET platelets were 8-16 times more sensitive to autoantibody and alloantibody, but 8-16 times less sensitive to d...

Antibody-dependent cell-mediated cytotoxicity (ADCC) toward human O+ red cells coated with anti-D antibody: comparison between lymphocyte and monocyte ADCC activity.

We investigated the antibody dependent cell-mediated cytotoxicity (ADCC) of lymphocytes and monocytes toward human O+ red cells coated with anti-D antibody using a 51Cr release assay. Lysis of sensitized red cells by lymphocytes occurred rapidly, but monocyte-mediated lysis occurred slowly. This difference might be due to postphagocytic 51Cr release by monocytes. ADCC of lymphocytes increased i...

A mechanism for neutrophil-mediated lysis of human neuroblastoma cells.

Neutrophils mediate the lysis of human neuroblastoma cells coated with human/mouse chimeric anti-GD2 ganglioside antibody ch14.18. This study examined the mechanism(s) by which this occurs. Neutrophil degranulation was found to be a required step for lysis, since release of granular enzymes from neutrophils correlated with the lysis of antibody-coated neuroblastoma cells. In addition, agents wh...

Effect of corticosteroid therapy on the phagocytosis of antibody-coated platelets by human leukocytes.

Patients with idiopathic thrombocytopenic purpura (ITP), a disorder in which antibody coated platelets are cleared from the circulation by phagocytic cells, are often treated with glucocorticoids. The effect of corticosteroids on the recognition and ingestion of sensitized platelets by phagocytes can be quantified in these patients and compared to changes in platelet levels. Six patients with I...
